Mechanism
Binds PTH1R on bone-building cells; given intermittently, it favors bone formation over resorption by activating Wnt/β-catenin signaling and suppressing sclerostin, boosting osteoblast activity and survival and raising local IGF-1 to support bone matrix production.
Safety file
The osteosarcoma boxed warning was removed in 2020; risks include high blood calcium and low blood pressure on standing; use is limited to two years.
