DSIP
Deltaran, Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu
A nine-residue peptide isolated in 1977 from sleeping rabbits. Mechanisms: GABA, adenosine, HPA/cortisol. Score 60/100, inconsistent clinical effect, and an FDA warning about antibody formation in compounded products.

- Circadian models
- Catalog fill
- 10 mg
- Typical water
- 2 mL
- Route
- Subcutaneous
- Cycle
- 1–2 weeks to assess, not an indefinite nightly peptide.
- Approval
- Research chemical only.
- Status
- Research chemical. FDA warning on compounded immunogenicity.
- Evidence
- Clinical efficacy described as inconsistent.
- Research score
- 60 / 100
Mechanism
Neuromodulator on GABAergic sleep regions, adenosine signalling, HPA-axis stress tone. Crosses the BBB in the models cited.
Safety file
Long-term human safety unknown. Grogginess and headache in anecdotes. FDA: potential life-threatening immunogenicity with compounded DSIP. Sedative interactions.

Reconstitution
2 mL bacteriostatic water yields 5 mg/mL, 50 mcg/unit. A 100 mcg evening amount is 2 units — easy to overshoot. Use a finer syringe if you have one.
Research amounts
Evening, 30–60 minutes before intended sleep.
| Step | Amount | Frequency | Notes |
|---|---|---|---|
| Common research amount | 100 mcg | Evening | 2 units from a 2 mL reconstitution |
| Upper cited amount | 300 mcg | Evening | 6 units |
Storage
- Lyophilized
- −20 °C freezer preferred; a refrigerator is also acceptable. Do not open a cold vial — wait 15–30 minutes so it does not sweat. Avoid freeze–thaw cycling.
- After water
- 2–8 °C, dark, up to 45 days when mixed with bacteriostatic water. Mixed vials stay in their box. Sterile water (no preservative) is single-use.
Worksheet notes
- Two units is a small draw. If 2 mL makes the amount unreadable, use 1 mL water instead (100 mcg = 1 unit at 10 mg/mL) only if you can measure 1 unit accurately.
- Bacteriostatic water: 20–25 °C. Do not refrigerate the diluent.
- Amounts on this sheet are reconstitution math for the vial, not a recommendation.
Full protocol
Titration, schedule, and handling
The complete protocol for DSIP. Where it differs from the worksheet amounts above, the worksheet reflects the NovaEvum vial.
Reconstitution
| Vial | 2 mL water | 3 mL water |
|---|---|---|
| 10 mg | 5 mg/mL | 3.33 mg/mL |
- Use bacteriostatic water as the diluent.
- Add the water slowly, letting it run down the inside wall of the vial rather than onto the powder.
- Swirl gently to mix, then leave it to dissolve. Do not shake.
- Give it a few minutes. The finished solution should be clear and colourless.
Protocol
Route: Subcutaneous
Standard protocol (subcutaneous)
- 0.1 mg to 0.2 mg (100 to 200 mcg), taken in the evening, ideally 1 to 3 hours before bed
- At a 2 mL fill on a 10 mg vial (5 mg/mL), 0.1 mg is a 0.02 mL draw (2 units)
Used at night, not daily forever
- DSIP is taken around sleep as needed
- Studies have run it up to about 14 days
- It is a sleep-timing tool, not an all-day peptide
Dosing detail
- Common dose is 0.1 mg to 0.2 mg, taken in the evening before bed.
- Delta sleep-inducing peptide, used for sleep quality.
- Inject at night, roughly 1 to 3 hours before sleep. Not dosed around the clock.
- Used as needed around sleep rather than on a fixed long cycle.
Duration
- Used at night as needed; study courses have run up to about 14 days.
Notes
- Delta sleep-inducing peptide, used for sleep quality.
- Inject in the evening.
- Inject subcutaneously and rotate sites.
Storage and handling
- Keep the powdered (lyophilized) vial in a cool, dark place. Refrigerate it for longer-term storage before mixing.
- Once reconstituted and refrigerated at 2–8 °C away from light, use the vial within 90 days.
- Do not freeze a reconstituted vial, and keep it away from heat and sunlight.
- If the solution turns cloudy, develops floating particles, or shows any other sign of degradation, stop using that vial.
