Orforglipron (LY3502970)
LY3502970, LY-3502970, Orforglipron
An oral, once-daily, non-peptide small-molecule GLP-1 receptor agonist for type 2 diabetes and obesity, offering a pill alternative to injected GLP-1 drugs. Trials through 2025 showed dose-dependent weight loss up to about 14.7% and better glucose control; it is now in Phase 3, including the ATTAIN weight-management program.
Research score 85/100. Reference only, not a dose.

Mechanism
Binds the GLP-1 receptor to raise cAMP and activate PKA, prompting glucose-dependent insulin release and less glucagon; also acts on hypothalamic neurons to cut appetite, slows stomach emptying via vagal signaling, and supports β-cell survival through PI3K/Akt.
Safety file
Nausea, vomiting, and diarrhea are common but usually mild and temporary; no liver-toxicity signal has emerged, unlike some earlier oral GLP-1 candidates.
