Mechanism
Irreversibly binds the β5 subunit of the 20S proteasome/immunoproteasome’s catalytic threonine, blocking chymotrypsin-like activity; this piles up ubiquitinated proteins, triggering ER stress, cell-cycle arrest, and apoptosis, especially in high-protein-turnover myeloma cells.
