Synthetic D-retro-inverso peptide (Proxofim) developed at Erasmus University Medical Center (2017) to selectively kill senescent (“zombie”) cells by disrupting FOXO4-p53 binding. Shown in mice to reduce age-related decline and support tissue function, especially in the brain; remains research-only with no human trials.

Mechanism
Blocks the FOXO4-p53 interaction that normally protects senescent cells from death, causing p53 to move to mitochondria and trigger apoptosis selectively in senescent cells; crosses the blood-brain barrier to target senescent neural tissue.

Human safety largely unestablished; theoretical risks include off-target apoptosis, immune activation, and metabolic disturbance, though animal models show good tolerability.
