PNC-27 / PNC-28
PNC27, PNC28, p53 peptide 27, p53 peptide 28, HDM-2 binding peptide
Synthetic peptides derived from p53 designed to bind HDM-2 on cancer cells and trigger pore formation and cell death. Preclinical data suggest selective cancer-cell killing, but there is no human clinical evidence, and the FDA has warned against their use.

Mechanism
A cell-penetrating domain lets these peptides reach HDM2 overexpressed on cancer-cell membranes, where binding causes pore formation and necrotic death rather than working through intracellular p53 signaling as typical p53-based therapies do.
Safety file
Human safety is unknown; selectivity claims come only from preclinical work, and the FDA has warned of contamination risks with sold products.
- Immune system activation
- Local injection site reactions
- Potential cytokine responses
